Many many MOC versions of Thomas and his friends already exist for 9v and

taking the Duplo Thomas parts and reusing them to build a system scale

Yes, with these, we'll have the correct LEGO elements necessary to pull it off.
People were doing ATSF warbonnets and BNSF units long before the official sets
came out, but their appearance made things a lot easier for kitbash purposes. In
this case I predict that purists especially will be more than happy to be using
ABS (albeit Duplo) in lieu of faces and other parts ripped off from other toys,
or having to build them in the brick. I see some great Thomas and friends MOCs
coming! I can't wait. :-)
